It would help if I knew whether your computer was recognizing the iPhone. When you connect the two, does your computer's operating system recognize it, and offer you the option to open it as a USB mass storage device? Does it show up in 'My Computer'?

Without answers to the above, I'd tend to suspect either (a) a communication issue between your iPhone and iTunes, or (b) an issue with the USB port you're using.

To check (b), try connecting the iPhone to the computer using a different USB port. Connect by plugging the dock cable directly into the port on the PC, without going through any external hubs, etc. if no change, check in Device Manager (winbloze) to ensure that the USB port is functioning normally.

After you've ruled out hardware issues, look at the software side. Update to the latest version of iTunes, and try again. If no joy, try this: With the phone connected to the computer, power it (the iPhone) off. Then restart the computer. As the computer starts up, it will power up the USB ports, sending power to the iPhone and turning it on. This will often clear up device recognition and hardware communication issues.
